Why can't they generate the assignments first and double-assign the unmatchable writer if/when they reply?

Best case scenario, unmatchable writers might take preemptively take care of a few initial pinch hits.

Also, if the unmatchable (offer) doesn't respond and their sign-up has to be deleted, they'll need to reassign the person who was assigned to write for them to someone else.

There are probably other reasons too.

You don't need to assign them a writer, they can be sent out together with the initial pinch-hits after they respond with a matchable offer.

That is a lot more work for mods than just waiting 24 hours for them to respond. Don't worry! Assignments will be sent out soon!

The writer who ao3 automatically assigned has to be assigned to someone else, so what you're suggesting increases the double-assignments and the initial pinch hits.

It's not a case of "matching is done and there's no one left for this person to write for," it's "there's no one POSSIBLE for this person to write for." Whatever fandom/character combinations they offered did not match up with anyone's requests at all. So they have to either expand their offers or dro out.

If the problem is just that all of a person's possible recipients are taken by other writers, the mods will assign them to someone they're compatible with who already has a writer; nobody gets asked about that. They only need to contact people who literally can't write for anyone. (People with no possible writers go out on the pinch hit list a few hours after assignments go out, with the super-early defaulters.)

If someone has chosen characters for a fandom in which all the requests have chosen other/more/different combination of characters, do the mods tell them that or are they just expected to choose different fandoms from scratch?

Well, presumably, you would know from the signup summary that your fandom has requests so you would know what fandoms you could change characters on.

Last year one of the unmatchable writers showed up on coal. IIRC, they said the mods provided a list of characters that will make them matchable on their offered fandoms, and gave them the option of either choosing from that list and/or offering new fandoms.
